The research at the Department of Biomedical Sciences of Cells & Systems (BSCS) at the UMC Groningen focusses on understanding basic functions and processes of the cell & systems and how they lead to diseases if disturbed. The Department strives for identification and, if possible, exploitation of targets to prevent or cure disease, and to promote Healthy Aging. The position will also be embedded in the MS Center Noord Nederland
Multiple sclerosis (MS) is a chronic progressive, disabling demyelinating, neuroinflammatory and neurodegenerative disorder of the central nervous system. The quiescence of oligodendrocyte progenitor cells is one of the major impediments to efficient remyelination in MS. Pathological amyloid-like extracellular fibronectin aggregates play a causative role in remyelination failure. We have recently shown that the underlying mechanism of fibronectin aggregation involves disrupted fibronectin fibrillogenesis and substantial protein conformational regulation likely regulated by extracellular HSP90ß. In this project you will examine in detail the requirements of HSP90ß in preventing accumulation of fibronectin aggregates and elucidate its dysfunction in MS that may encompass aberrant post-translational modifications. The project will result in the identification and validation of an extracellular HSP90ß targeting agent that selectively prevents accumulation of fibronectin aggregates.
The position is funded by Stichting MS Research.
